We've rapidly generated Schwann cell precursors (SCP) from human induced pluripotent stem cells (hiPSCs) in just 9 days, offering a promising model for studying diseases like diabetic neuropathy and Charcot-Marie-Tooth syndrome. These SCPs, when co-cultured with our hiPSC-derived sensory and motor neurons, align within a week. Myelination markers are expressed as the SCPs are matured and further validated by transmission electron microscopy. Our model allows for screening with tool compounds and functional analysis using multi-electrode arrays, advancing the study of peripheral nervous system diseases.

Comparative PCA Analysis of RealSCP and Primary Schwann Cell Populations
Bulk RNA sequencing of the RealSCP population was conducted and compared to multiple published datasets, including other hiPSC-derived Schwann cells and primary Schwann cell populations. The data indicates total days in vitro for each cell type. Principal component analysis (PCA) shows that the RealSCP population clusters closely with primary human Schwann cell datasets at both day 9 in vitro (0 days of maturation) and day 16 (7 days of maturation). in vitro (7 days of maturation), highlighted in red.
